- Antibody CD55-PE is a monoclonal antibody (mAb) labelled with R-phycoerythrin (PE) designed for flow cytometry (FC) use as a direct immunofluorescence reagent in the identification and enumeration of CD55 antigen-expressing cells.
SUMMARY AND EXPLANATION
FC is a powerful tool in analytical and quantitative characterization of cells which provides rapid and multiparametric analysis of heterogeneous cell populations on a cell-by-cell basis. Flow cytometry is performed on cell suspension after incubating it with fluorescentlabelled antibodies directed against specific cellular proteins. Positive cells relative fluorescence intensity indicates the amount of antibody bonded to specific cell sites providing information about antigen expression. Several key complement regulatory proteins regulate the activation of complement cascade preventing damage to autologous cells during an inflammatory reaction. Complement regulatory protein CD55 works down-regulating the complement cascade at the critical step of C3 and C5 activation. CD55 is expressed in all haematopoietic cells that circulate in contact with plasma complement proteins as well as on a wide spectrum of cells of non-haematopoietic origin and its overexpression was observed in several malignant tumour cells stopping them from being destroyed by complement activity. In paroxysmal nocturnal haemoglobinuria (PNH) GPI tail proteins domain have a synthetic defect which leads to a deficient expression of all GPI-anchored proteins (e.g. CD55 and CD59). For that reason, CD55 together with CD59 have been recurrently used in PNH diagnosis.

The purified monoclonal CD55 Antibody conjugated with R-phycoerythrin (PE) is supplied in phosphate-buffered saline (PBS) containing 0.1% sodium azide.
