Description and applications: This anti-hepatitis C virus mouse monoclonal antibody recognises an epitope NS3P of 70kDa of HCV that is stable to routine processing and exhibits minimal crossreactivity with normal or diseased liver, although non-specific staining has been observed in heavily immunosuppressed individuals with cholestasis. The antibody staining is positive in most confirmed cases of hepatitis C, in more than half presenting intense cytoplasmic positivity. The antibody is also useful for identifying primary HCV infection in liver biopsies as antigen expression is present even in patients with no serologically apparent infection. However cases with more intense positivity are those with higher viral load and more severe liver injury. A decrease of protein expression and HCV replication is observed during tumor progression therefore hepatocarcinomas related to viral infection confirmed by RT-PCR techniques may show negative staining with this antibody. However, persistent expression in the hepatocytes surrounding dysplastic nodules, suggests an active role of HCV in carcinogenesis.v.
