SMAD4 is a protein that is involved in cell signaling. It functions as a transcription factor and is a member of the TGFbeta superfamily. It is mutated in many cancers: multiploid colorectal cancer, pancreatic carcinoma and juvenile polyposis syndrome (JPS) which includes a greater risk of developing gastrointestinal cancers, in particular colon cancer. SMAD4 is a member of the Mothers Against Dpp (MAD)-related family of proteins.
