PAX8 antibody recognizes a protein of 62kDa, identified as PAX8. It is a member of the paired box (PAX) family of transcription factors. This nuclear protein is involved in thyroid follicular cell development and expression of thyroid specific genes. Mutations in this gene have been associated with thyroid dysgenesis, thyroid follicular carcinomas, and atypical thyroid adenomas. PAX8 is expressed in the thyroid (and associated carcinomas), non-ciliated mucosal cells of the fallopian tubes, and simple ovarian inclusion cysts, but normal ovarian surface epithelial cells. Clone 100/D5 reacts with bcl-2 alpha oncoprotein as assessed by reaction with a 26-kD band in immunoblotting. This antibody stains the 380 cell line which carries a 14;18 chromosomal translocation. bcl-2 oncogene codes for two closely similar proteins, bcl-2 alpha and bcl-2 beta. These two proteins differ only in their carboxyl terminal sequences. bcl-2 is an integral inner mitochondrial membrane protein and is frequently overexpressed in many lymphoid malignancies. β-Catenin is a 92 kDa protein that binds to the cytoplasmic tail of E-Cadherin. The cadherins, transmembrane adhesion molecules, are found with catenin’s at adherens junctions (zonula adherens). Deletions in the cytoplasmic domain of E Cadherin which eliminate catenin binding also result in a loss of cell adhesion, indicating that this binding is essential for E Cadherin function. Although the α- and β-Catenin’s have been cloned, very little is known about their biochemical roles. However, a link between β-Catenin and colon cancer has been described. β-Catenin was found to co-immunoprecipitated with the APC tumor suppressor protein in human colorectal tumor cell lines, as well as in human kidney 293 cells. E-Cadherin, however, was not detectable in these complexes. Thus, the APC Catenin complex may be affecting the transmission of contact inhibition signals and/or the regulation of cell adhesion.

CEA (carcinoembryonic antigen, CD66) consists of a heterogeneous family of related oncofetal 200 kD glycoproteins that is secreted into the glycocalyx surface of gastrointestinal cells. The staining of CEA is considered important because: 1. In breast carcinomas, CEA staining correlated well with clinical outcome, independent of histologic type of tumors (with or without metastases). 2. A CEA-staining primary tumor usually shows CEA elevations when there is disease recurrence or progression. 3. Before proceeding with extensive or repeated blood monitoring of CEA as an index of biological activity of diseases, it is important to find out if the substance is present in the original malignant lesion, preferably at the time of biopsy. 4. Usually CEA is demonstrated as a linear labeling of the apical poles of cells lining the glandular lumen and, occasionally, as weak staining near the apex of colonic epithelial cells. CEA, however, should not be used as a marker of differentiation because many colon and lung tumors actually show increased staining with differentiation. Pancreatic carcinomas, testicular tumor, gall bladder neoplasms and granular cell Myo blastomas stain positive, whereas malignant tumors of brain, prostate, skin, lymphoreticular tissues, hepatocellular carcinomas, esophageal squamous cell carcinomas, and mesothelioma fail to stain for CEA.

Factor 13 (plasma transglutaminase, fibrin stabilizing factor) is a plasma protein that plays an important role in the final stages of blood coagulation and fibrinolysis. It circulates in blood as a tetramer consisting of two “A” and two “B” subunits.
