SUMMARY AND EXPLANATION
CD33 antigen is a 67 kD type I transmembrane glycoprotein restricted to myelomonocytic lineage. CD33 is expressed on monocytes and macrophages, granulocytes, myeloid progenitors (not the earliest precursors), and myeloid dendritic cells. It is absent on normal platelets, lymphocytes, erythrocytes, and hematopoietic stem cells. CD33 expression is brighter in monocytes and dimmer in mature granulocytes (expression decreases with maturation and differentiation). CD33 antibody is used in flow cytometry for identifying and immunophenotyping myeloid leukemias. It is a high valuable antigen to distinguish acute myeloid leukemias from their lymphoblastic and erythroblastic counterparts. It is also a key marker to diagnose myelodysplastic syndromes by detecting dysplasia in the different myeloid compartments.
REAGENT COMPOSITION
The purified monoclonal CD33 Antibody conjugated with allophycocyanine (APC) is supplied in phosphate-buffered saline (PBS) containing 0,09% sodium azide.
