- Antibody CD43-FITC is a monoclonal antibody (mAb) labelled with fluorescein isothicyanate (FITC) designed for use as a direct immunofluorescence reagent in the identification and enumeration of cells which express the CD43 antigen by Flow Cytometry (FC).
SUMMARY AND EXPLANATION
FC is a powerful tool for the analytical and quantitative characterization of cells which provides rapid, quantitative and multiparametric analysis of heterogeneous cell populations on a cell-by-cell basis. FC is performed on cells in liquid suspension that have been incubated with fluorescently-labeled antibodies directed against specific cellular proteins. The relative fluorescence intensity of the positive cells indicates the amount of antibody bound to specific binding sites on the cells, and therefore provides a relative measure of antigen expression. CYT-43F2 mAb recognizes the CD43 antigen, also known as Ly-48, Leukosialin, Sialophorin, Leukocyte Sialoglycoprotein and W3/13, is a large single chain of type I transmembrane glycoprotein with abundant O-glycosylation and sialylation sites. CD43 exerts dual function in cell adhesion and anti-adhesion, lectin binding, as well as co-stimulation of T cell activation and survival, and induction of apoptosis of T cells and hematopoietic progenitors .During T-cell activation is actively removed from the T-Cell APC (antigen presenting cell) contact site thus suggesting a negative regulatory role in adaptive immune response. The expression of CD43 is found on most leukocytes, except resting B lymphocytes.
