    Specification:

    p63 is a homolog of the tumor suppressor p53. It is identified in basal cells in the epithelial layers of a variety of tissues, including epidermis, cervix, urothelium, breast and prostate. p63 was detected in nuclei of the basal epithelium in normal prostate glands; however, it was not expressed in malignant tumors of the prostate. As a result, p63 has been reported as a useful marker for differentiating benign from malignant lesions in the prostate, particularly when used in combination with markers of high molecular weight cytokeratin’s and the prostate-specific marker AMACR (P504S clone 13H4). p63 has also been shown to be sensitive marker for lung squamous cell carcinomas (SCC), with a sensitivity of ~90%. Specificity for lung SCC, vs. lung adenocarcinoma (LADC), is approximately 80%. In breast tissue, p63 has been identified in myoepithelial cells of normal ducts.

    Recognizes a protein of 55kDa, identified as CD4. It is a membrane glycoprotein of T lymphocytes that interacts with major histocompatibility complex class II antigens and is also a receptor for the human immunodeficiency virus.     Giardia lamblia is a flagellated protozoan parasite that infects mammalian gastrointestinal tracts and causes giardiasis, a type of gastroenteritis that manifests itself with severe diarrhea and abdominal cramps. Giardia lamblia infection is transmitted by consuming contaminated food or water, or through mammalian feces. The Giardia protozoan cannot survive in an environment outside of the host, so it is spread as a cyst, which can live in the environment for up to a month and enters its trophozoite active form after infection.     ZAP-70 is a 70kDa protein tyrosine kinase found in T-cells and natural killer cells. Control of this protein translation is via the IgVH gene. In Western blotting of whole cell lysates of normal peripheral blood mononuclear cells, the antibody labels a band corresponding to ZAP-70. In Western blotting of whole cell lysates of CD19-positive purified leukemia cells from patients with lg-unmutated and lg-mutated CLL, the antibody labels a band corresponding to ZAP-70 in the Ig-unmutated CLL samples, whereas no band is observed in the Ig-mutated CLL samples. In Western blotting of cell lysates of Jurkat cells (T-lymphoblastic cell line), the antibody labels a band of 70kDa protein. In Western blotting of cell lysates of A431 cells (carcinoma cell line), no band is observed. ZAP-70 protein is expressed in leukemic cells of approximately 25% of chronic lymphocytic leukemia (CLL) cases as well. Anti-ZAP-70 expression is an excellent surrogate marker for the distinction between the g-mutated (antiZAP-70 negative) and Ig-unmutated (anti-ZAP-70 positive) CLL subtypes and can identify patient groups with divergent clinical courses. The anti-ZAP-70 positive Ig-unmutated CLL cases have been shown to have a poorer prognosis.

    Originally, the 2G9 monoclonal antibody (mAb) was described as identifying a 15 kDa protein found in the cytoplasmic granules of cytotoxic T cells that might be part of a larger 40 kDa molecule, ubiquitously expressed, named p40-TIA-1 and often referred to as TIA-1 in the literature. Now, however, there is evidence that the 2G9 mAb identifies a 17 kDa cytoplasmic granule membrane protein named GMP-17 that has no similarity with p40-TIA-1. The GMP-17 antigen is a 165 amino acid protein with 4 transmembrane domains: but it is not a typical member of the four transmembrane superfamily. It is identical with previously identified cytotoxic granule proteins called NKG7 and GIG-1 – for GCSF induced gene protein 1 –, isolated from NK cells and granulocyte-colony stimulating factor-treated mononuclear cells, respectively.  The GMP-17 protein is also found in the granules of CD14+ monocytes and neutrophils. Conversely, GMP-17 is not expressed in B lymphocytes or B-cell lines. In humans, NKG7, the GMP-17 gene, is localized on chromosome 19q13-33. As the target cell-induced NK cell degranulation results in translocation of GMP-17 from granules to the plasma membrane, a possible role for GMP-17 in the formation of junctions between effector cells and target cells has been suggested. Furthermore, sequence homology with calcium channel proteins has suggested that it may regulate ion channels required for cytotoxic effector functions. The 2G9 mAb was evaluated during the 5th HLDA Workshop on Human Leucocyte Differentiation Antigens, in the section of monoclonal antibodies reactive with intracellular antigens.

    Membrane receptor signaling by various ligands, including interferons and growth hormones such as EGF, induces activation of JAK kinases which then leads to tyrosine phosphorylation of proteins that have been designated Stats (signal transducers and activators of transcription). The first members of this family to be described include Stat1α p91, Stat1β p84 (a form of p91 that lacks 38 COOH-terminal amino acids) and Stat2 p113. Stat1 and Stat2 are induced by IFN-α and form a heterodimer which is part of the ISGF-3 transcription factor complex. Stat3, which becomes activated in response to epidermal growth factor (EGF) and interleukin-6 (IL-6), but not interferon-γ (IFN-γ) or Stat4, is an additional member of this family. It has been suggested that the phosphorylated forms of both Stat3 and Stat4 form homodimers as well as heterodimers with the other members of the Stat family, and that differential activation of different Stat proteins in response to different ligands should help to explain specificity in nuclear signaling from the cell surface. Highest expression of Stat4 is seen in testis and myeloid cells. IL-12 has been identified as an activator of Stat4. Other members of the Stat family include Stat5, which has been shown to be activated by Prolactin and by IL-3, and Stat6 (also designated IL-4 Stat), which is involved in IL-4-activated signaling pathways.

    Cytokeratin 14 (CK14) belongs to the type I (or A or acidic) subfamily of low molecular weight keratins and exists in combination with keratin 5 (type II or B or basic). CK14 is found in basal cells of squamous epithelia, some glandular epithelia, myoepithelium, and mesothelial cells. Anti-CK14 is useful in differentiating squamous cell carcinomas from poorly differentiated epithelial tumors. Anti-CK14 is one of the specific basal markers for distinguishing between basal and nonbasal subtypes of breast carcinomas.     Hepatocyte Specific Antigen, also called Hepatocyte Paraffin 1 or Hep Par 1, localizes to the mitochondria of hepatocytes. It is a sensitive marker for distinguishing hepatocellular carcinomas (HCC) from other metastatic carcinomas as well as cholangio-carcinomas.     TDT is a DNA polymerase that catalyses the addition of deoxynucleotides to free 3’OH groups on polydeoxynucleotide chains. TDT is present in the nucleoli of normal T and B lymphocyte precursors and the neoplastic equivalent. Positive cells are most abundant in normal thymus, especially in the cortex, with little or no labelling of the medulla. A few cells in the normal bone marrow corresponding to hematopoietic precursor cells, also express TDT. TDT is a valuable marker in the identification of immature T and B Lymphocyte precursors and acute leukaemia’s.     Myogenin is a member of the MyoD family of myogenic basic helix-loop-helix (bHLH) transcription factors that also includes MyoD, Myf-5, and MRF4 (also known as herculinor Myf-6). MyoD family members are expressed exclusively in skeletal muscle and play a key role in activating myogenesis by binding to enhancer sequences of muscle-specific genes.     CD8 is a cell surface receptor expressed either as a heterodimer with the CD8 beta chain (CD8 alpha/beta) or as a homodimer (CD8 alpha/alpha). A majority of thymocytes and a subpopulation of mature T cells and NK cells express CD8a. CD8 binds to MHC class 1 and through its association with protein tyrosine kinase p56lck plays a role in T cell development and activation of mature T cells. For mature T-cells, CD4 and CD8 are mutually exclusive, so anti-CD8, generally used in conjunction with anti-CD4.
