Specification:

Recognizes a 67kDa transmembrane protein, which is identified as CD5. The CD5 antigen is found on 95% of thymocytes and 72% of peripheral blood lymphocytes. In lymph nodes, the main reactivity is observed in T cell areas. CD5 is expressed by many T cell leukemia, lymphomas, and activated T cells. Occasionally, CD5 antigen is also expressed on a subset of B cells. Mantle cell lymphomas (same as diffuse centrocyte lymphomas) are CD5+ while the follicle center cell lymphoma is CD5-negative. This MAb is superb for identifying the formalin-fixed, paraffin-embedded mantle cell lymphomas.

CD34 monoclonal antibody recognizes a cell surface antigen of approximately 110Kd that is expressed selectively on human hematopoietic progenitor cells, including myeloid and lymphoid lineage progenitors, and a significant portion of acute leukemia. In addition to this stem cell recognition, this marker is expressed by vascular endothelium. Additionally, it appears that proliferating endothelial cells express this molecule in greater amounts than non-proliferating endothelial cells.

Recognizes a protein of 185kDa, which is identified as c-erbB-2/HER-2/neu. Its epitope is localized in the extracellular domain. C-erbB-2/HER-2 is a member of the EGFR family. This MAb is specific and shows minimal cross-reaction with other members of the EGFR-family. Receptors of this family are located on the plasma membrane and consist of an extracellular ligand-binding domain that is connected to a large intracellular domain by a single transmembrane sequence. c-erbB 2/HER-2 protein is over-expressed in a variety of carcinomas especially those of breast and ovary.

This antibody recognizes an intermediate filament protein (IFP) of 55kDa, which is identified as cytokeratin 7. This MAb is highly specific to cytokeratin 7 and shows no cross-reaction with other IFPs. Cytokeratin 7 is a basic cytokeratin, which is found in most glandular and transitional epithelia but not in the stratified squamous epithelia. Keratin 7 is expressed in the epithelial cells of ovary, lung, and breast but not of colon, prostate, or gastrointestinal tract. This MAb is highly useful in distinguishing ovarian carcinomas (keratin 7+) from colon carcinomas (keratin 7-).

β-Catenin is a 92 kDa protein that binds to the cytoplasmic tail of E-Cadherin. The cadherins, transmembrane adhesion molecules, are found with catenin’s at adherens junctions (zonula adherens). Deletions in the cytoplasmic domain of E Cadherin which eliminate catenin binding also result in a loss of cell adhesion, indicating that this binding is essential for E Cadherin function. Although the α- and β-Catenin’s have been cloned, very little is known about their biochemical roles. However, a link between β-Catenin and colon cancer has been described. β-Catenin was found to co-immunoprecipitated with the APC tumor suppressor protein in human colorectal tumor cell lines, as well as in human kidney 293 cells. E-Cadherin, however, was not detectable in these complexes. Thus, the APC Catenin complex may be affecting the transmission of contact inhibition signals and/or the regulation of cell adhesion.

It recognizes a protein of about 29kDa, which is identified as Calretinin (also known as Calbindin 2). Calretinin is a vitamin D dependent calcium-binding protein involved in calcium signaling. It is present in subsets of neurons throughout the brain and spinal chord, including sensory ganglia. Antibody to calretinin is useful in differentiating mesothelioma from adenocarcinomas of the lung. It also aids in differentiating adrenal cortical neoplasms from pheochromocytomas.

CD19 is a transmembrane glycoprotein that contains two extracellular immunoglobulin-like domains. CD19 is present in both benign and malignant B-cells and is considered to be the reliable surface marker of this lineage over a wide range of maturational stages. In normal lymphoid tissue, CD19 is observed in germinal centers, in mantle zone cells and in scattered cells of the inter-follicular areas. Anto-CD19 exhibits an overall immunoreactivity pattern similar to those of the antibodies against CD20 and CD22. However in contrast to CD20, expression of CD19 is continuous throughout B-Cell development and trough terminal differentiation of B-cells into plasma cells. Anti-CD19 positivity is seen in the vast majority of B-cells neoplasms commonly at a lower intensity than normal B-cell counterparts. Plasma cell neoplasm are nearly always negative, as are t-cells neoplasms.

Cytokeratins 8 & 18 can be found in most simple epithelium, e.g. thyroid, female breast, gastrointestinal tract, and respiratory tract. Adenocarcinomas and most non-keratinizing squamous carcinomas will stain but keratinizing squamous carcinomas will not. This antibody is used when attempting to demonstrate the presence of Paget cells; there is very little keratin 18 in the normal epidermis so this will only stain Paget cells. This approach facilitates the interpretation using immunostains and is more sensitive than mucin histochemistry.

Specification:

Anti-B72.3 recognizes a high molecular weight glycoprotein called TAG-72 that is present in human adenocarcinomas and in lesser amounts, non-neoplastic tissues. Specification:

CD31 (PECAM-1) is a transmembrane glycoprotein member of the immunoglobulin supergene family of adhesion molecules. It is widely used as an endothelial cell marker. CD31 Ab reacts with normal, benign, and malignant endothelial cells which make up blood vessel lining. The level of CD31 expression can help to determine the degree of tumor angiogenesis, and a high level of CD31 expression may imply a rapidly growing tumor and potentially a predictor of tumor recurrence.
