Name: Flow Cytometry Antibody CD33-APC, Clone WM53

    • Antibody CD33-APC is a monoclonal antibody (mAb) labelled with allophycocyanine (APC) designed for flow cytometry (FC) use as a direct immunofluorescence reagent in the identification and enumeration of CD33 antigen-expressing cells. This reagent must be used by FC qualified personal.

    SUMMARY AND EXPLANATION

CD33 antigen is a 67 kD type I transmembrane glycoprotein restricted to myelomonocytic lineage. CD33 is expressed on monocytes and macrophages, granulocytes, myeloid progenitors (not the earliest precursors), and myeloid dendritic cells. It is absent on normal platelets, lymphocytes, erythrocytes, and hematopoietic stem cells. CD33 expression is brighter in monocytes and dimmer in mature granulocytes (expression decreases with maturation and differentiation). CD33 antibody is used in flow cytometry for identifying and immunophenotyping myeloid leukemias. It is a high valuable antigen to distinguish acute myeloid leukemias from their lymphoblastic and erythroblastic counterparts. It is also a key marker to diagnose myelodysplastic syndromes by detecting dysplasia in the different myeloid compartments.

    REAGENT COMPOSITION

The purified monoclonal CD33 Antibody conjugated with allophycocyanine (APC) is supplied in phosphate-buffered saline (PBS) containing 0,09% sodium azide.

    • Clone: WM53.

Name: Flow Cytometry Antibody CD33-FITC, Clone WM53

    • CD33-FITC is a monoclonal antibody (mAb) labelled with fluorescein isothiocyanate (FITC) designed for use as a direct immunofluorescence reagent in the identification and enumeration of cells which express the CD33 antigen by flow cytometry (FC). Anti-CD33 is considered essential for the initial evaluation of acute myeloid leukemia (AML) together with a panel of other antibodies. This reagent must be used by FC qualified personal.

    SUMMARY AND EXPLANATION

The CYT-33F mAb reacts with a member of the siglec family of 67kDa expressed by peripheral blood monocytes, neutrophils, granulocyte precursors, myeloid progenitor cells and cells of myeloid leukemias. Analysis of the expression of antigen CD33 is useful for the identification and the characterization of hematologic disorders since CD33 has been found on the cell surface of leukemic blasts from the majority of cases of AML. Leukemic lymphoblasts do not show any expression of this antigen.

Name: Flow Cytometry Antibody CD38-APC-C750™, Clone LD38

    • Antibody CD38 is a monoclonal antibody (mAb) designed for flow cytometry (FC) use as direct (or indirect for the CD38-PURE) immunofluorescence reagent for the identification and enumeration of CD38 antigen-expressing cells. This reagent must be used by flow cytometry qualified personnel.

    SUMMARY AND EXPLANATION

CD38 antigen is a type II transmembrane glycoprotein expressed at variable levels on the majority of hematopoietic cells (during early differentiation and activation) and in some non-hematopoietic tissues. It is expressed at high levels on plasma cells (CD38+++) and, due to its strong reactivity, CD38 is considered a “specific plasma cell marker”; but it is also present on thymocytes and activated T cells. CD38 has been reported to play a role as regulator (positive and negative) of cell activation and proliferation, and to be involved in adhesion between lymphocytes and endothelial cells. CD38 antibody is used in FC for diagnosis and monitoring of plasma cell disorders (e.g. multiple myeloma, MGUS, plasma cell leukemia, etc.) .The simultaneous assessment of the expression of CD38 and CD138 represents the best combination of markers for the identification of plasma cells. Plasma cells (CD38+++) are expressed as a percentage of leucocytes present in a sample. Since every flow cytometer has different operating characteristics each laboratory must determine its optimal operating procedure.

    REAGENT COMPOSITION

CD39 is a cell surface protein member of the ecto-nucleoside triphosphate diphosphohydrolase (E-NTPDase) family. It is a 78 kDa cell surface enzyme also known as ectonucleoside triphosphate diphosphohydrolase 1 (ENTPD1). CD39 is expressed by regulatory T cells, mantle zone B cells, activated T cells, NK cells, macrophages, dendritic cells, neurons, endothelial cells and platelets. It is thought that CD39 allows Treg cells to enter inflamed areas where high levels of ATP are present. In the B-CLPD (B-cell chronic lymphoproliferative diseases) EuroFlow panel, CD39 is used to improve differential diagnosis between subgroups of lymphoplasmacytic lymphoma, marginal zone lymphoma, follicular lymphoma and diffuse large B-cell lymphoma cases.

    SUMMARY AND EXPLANATION

FC is a powerful tool for the analytical and quantitative characterization of cells which provides rapid, quantitative and multiparametric analysis of heterogeneous cell populations on a cell-by-cell basis. FC is performed on cells in liquid suspension that have been incubated with fluorescently labeled antibodies directed against specific cellular proteins. The relative fluorescence intensity of the positive cells indicates the amount of antibody bound to specific binding sites on the cells, and therefore provides a relative measure of antigen expression. CYT-43AC750 mAb recognizes the CD43 antigen, also known as Ly-48, Leukosialin, Sialophorin, Leukocyte Sialo glycoprotein and W3/13, is a large single chain of type I transmembrane glycoprotein with abundant O-glycosylation and sialylation sites. CD43 exerts dual function in cell adhesion and anti-adhesion, lectin binding, as well as costimulation of T cell activation and survival, and induction of apoptosis of T cells and hematopoietic progenitors .During T-cell activation is actively removed from the T-Cell APC (antigen presenting cell) contact site thus suggesting a negative regulatory role in adaptive immune response. The expression of CD43 is found on most leukocytes, except resting B lymphocytes. APC-C750TM is a tandem dye with a maximum emission peak at 779nm, which grants bright signal, low unspecific noise and high photostability. When excited by light from a red laser, the APC fluorochrome can transfer energy to C750 molecule, which then emits at a longer wavelength. It is recommended to use a 780/60 nm bandpass filter along with a red sensitive detector to use in conjunction antibodies conjugated with APC and APC-C750TM .

Leukocytes are named according to structure as being either granulocytes or agranulocytes, and according to the function as either phagocytes or immunocyte. The granulocytes, which include neutrophils, basophils and eosinophils, are all phagocytes. Of the agranulocytes, the monocytes and macrophages are phagocytes whereas the lymphocytes are immunocytes. The granulocytes have granules in their cytoplasm, which contain enzymes which are capable of killing microorganisms and destroying debris ingested by the process of phagocytosis. The CYT-45AP5 mAb reacts with the leucocyte common antigen (LCA) complex, which is present on lymphocytes, monocytes, granulocytes, eosinophils, basophils and their progenitors. This reagent can be used in the characterization studies for immunophenotyping of leucocytes, which are widely applied in the characterization and follow-up of immunodeficiencies, autoimmune diseases, leukemias, etc.

The anti-CD45 mAb reacts with the leukocyte common antigen (LCA) complex, which is present on lymphocytes, monocytes, granulocytes, eosinophils, basophils and their progenitors. This reagent can be used in the characterization studies for immunophenotyping of leukocytes, which are widely applied in the characterization and follow-up of immunodeficiencies, autoimmune diseases, leukemias, etc.

Name: Flow Cytometry Antibody CD45-OC515TM, Clone GA90

    • Antibody CD45-OC515TM is a monoclonal antibody (mAb) labelled with the fluorochrome Orange Cytognos 515 and designed for use as a direct immunofluorescence reagent in the identification and enumeration of cells which express the CD45 antigen by flow cytometry (FC).

    SUMMARY AND EXPLANATION

FC is a powerful tool for the analytical and quantitative characterization of cells which provides rapid, quantitative and multiparametric analysis of heterogeneous cell populations on a cell-by-cell basis. FC is performed on cells in liquid suspension that have been incubated with fluorescently-labeled antibodies directed against specific cellular proteins. The relative fluorescence intensity of the positive cells indicates the amount of antibody bound to specific binding sites on the cells, and therefore provides a relative measure of antigen expression. The CYT-45OC mAb reacts with the leucocyte common antigen (LCA) complex, which is present on lymphocytes, monocytes, granulocytes, eosinophils, basophils and their progenitors. This reagent can be used in the characterization studies for immunophenotyping of leucocytes, which are widely applied in the characterization and follow-up of immunodeficiencies, autoimmune diseases, leukemias, etc. Orange Cytognos 515 is excited with the violet laser (405nm) and emits at 515nm. This fluorochrome provides maximum resolution and narrow emission peaks, which results in little spectral overlap.

    SUMMARY AND EXPLANATION

Leukocytes are named according to structure as being either granulocytes or agranulocytes, and according to the function as either phagocytes or immunocyte. The granulocytes, which include neutrophils, basophils and eosinophils, are all phagocytes. Of the agranulocytes, the monocytes and macrophages are phagocytes whereas the lymphocytes are immunocytes. The granulocytes have granules in their cytoplasm, which contain enzymes which are capable of killing microorganisms and destroying debris ingested by the process of phagocytosis. The CYT-45PBZ mAb reacts with the leucocyte common antigen (LCA) complex, which is present on lymphocytes, monocytes, granulocytes, eosinophils, basophils and their progenitors. This reagent can be used in the characterization studies for immunophenotyping of leucocytes, which are widely applied in the characterization and follow-up of immunodeficiencies, autoimmune diseases, leukemias, etc. Pacific BlueTM is excited with the violet laser (405nm) and emits at 550nm. This fluorochrome provides maximum resolution and narrow emission peaks, which results in little spectral overlap and minimal compensation requirements. In multicolor panel it is recommended the use of this fluorochrome combined with Orange Cytognos 515, which is also excited with the violet laser and emits at 515nm.
